“Prospects for Multi-Focal Motor Neuropathy Treatment by Complement Inhibition”

Multifocal Motor Neuropathy (MMN) remains a challenging immune-mediated condition with limited durable treatment options beyond IVIG. Our review highlights the emerging therapeutic role of complement inhibition—targeting key components of the classical and lectin pathways involved in MMN pathogenesis.
💡 Key insights:
- Complement activation plays a role in MMN immune-mediated damage.
- C5 inhibitors like eculizumab show safety, but limited efficacy.
- Newer agents (e.g. ARGX-117, a C2 inhibitor) show early success in reducing IVIG reliance and improving function.
- Ongoing clinical trials (C2 and C1s inhibitors) are evaluating these novel therapies.
These findings may pave the way for disease-modifying treatment strategies in MMN and other immune neuromuscular diseases.
